摘要:
研究怀牛膝多糖(ABPS)对小鼠单核巨噬细胞RAW264.7细胞活性、表型及细胞因子表达的影响。先用不同浓度的ABPS溶液(0,0.4,0.6,0.8,1.0 mg/mL)处理RAW264.7细胞48 h,后采用MTT法、中性红吞噬实验、流式细胞术分别检测了ABPS对巨噬细胞的增殖活性、吞噬能力、细胞表型及细胞因子表达的影响。ABPS对RAW264.7细胞的增殖活性没有明显的影响,但能显著提高细胞对中性红的吞噬活性和表面分子CD80,CD86及CD40的表达,降低单核细胞趋化蛋白-1(monocyte chemotactic protein 1, MCP-1)和肿瘤坏死因子α(tumor necrosis factor α, TNF-α)的表达。ABPS能够增加RAW264.7细胞的吞噬活性并促进其成熟,且能发挥一定的抗炎活性。
Abstract:
To study the effect of Achyranthes bidentata polysaccharide(ABPS)on the activity, phenotype and cytokine expression of RAW264.7 cells(murine mononuclear macrophages). RAW264.7 cells were treated with different concentrations of ABPS solution(0, 0.4, 0.6, 0.8, 1.0 mg/mL)for 48 hours. Then, the effects of ABPS on the proliferation, phagocytosis, cell phenotype and cytokine expression of macrophage were analyzed by MTT assay, neutral red phagocytosis assay and flow cytometry respectively. ABPS had no significant effect on the proliferation of RAW264.7 cells, but significantly increased cell phagocytic activity and the expressions of CD80, CD86 and CD40. In addition, ABPS decreased the expressions of monocyte chemotactic protein 1(MCP-1)and tumor necrosis factor α(TNF-α). ABPS increased the phagocytic activity of RAW264.7 cells, promoted cell maturation, and exerted certain anti-inflammatory activity.
