摘要:
以盐酸氢氯噻嗪为模板分子, 以甲基丙烯酸甲酯(MAA) 为功能单体, 以三-羟甲基丙烷-三-丙烯酸酯为交联剂, 偶氮二异丁腈为引发剂, 并通过实验条件的优选, 合成了对氢氯噻嗪具有特异作用的分子印迹聚合物。通过测定印迹聚合物的 结合量, 评价了其对氢氯噻嗪的亲和力。S catchard 分析表明, 在印迹聚合物中存在对氢氯噻嗪形成不同亲合力的两类结合位 点, 这两类不同结合位点的离解常数分别为K d1 =57.0 μmol/ L 和K d2 =3.96 m mol/ L ;其最大表观结合量分别为Qmax 1 = 5.59 μm ol/ g 和Qmax 2 =216.4 μmol/ g 。
Abstract:
A synthetic polymer selective to Hydro chloro thiazide w as prepared by a non -covalent ly imprinted technique , using Hydrochlorothiazide as a template , methyl methacrylate as a functio nalmonomer and t ri - [ hy droxymethy l] -propan -tri -acrylic acid as a cross -linker , and methanol as a porog enic solvent .After extraction of template , affinity of the polymer imprinted with Hydro chloro thiazide w as examined in re-binding experiment .From the Scatchard analysis , binding site heterog eneity is obvious and two types of binding sites can be ident ified .The equilibrium dissociation co nstant K d1 , the apparent maximum amount Qmax1 and the binding constant Ka1 of the hig her aff inity binding sites can be calculated to be 57.0 μmo l/L , 5.59 μmo l/g , and 1.75 ×104 (mol/L)-1 ;w hile K d2 , Qmax2 , and K a2 of the lower affinity binding sites were calculated to be 3.96 mmol/L , 216.4 μmol/g and 2.53 ×102 (mo l/L)-1 , respectively .
