摘要:
以6 , 8 -二氯辛酸乙酯、硫化钠为起始原料, 甲醇和水作为溶剂合成中间产物硫辛酸乙酯, 水解酸化得外消旋α-硫辛 酸。外消旋产物利用R - (+) -α-甲基苄胺为拆分剂, 在甲苯溶剂中考察不同原料配比下ALA 的手性拆分效果, 确定了最 佳的拆分条件。结果表明:在反应温度40 ℃, 甲苯溶剂量30 mL , n (PEA) ∶n (ALA) =0.5 ∶1 的条件下, 进行非对映体 盐法拆分, 产物(R) - (+) -A LA 的收率达到53.0 %, 旋光纯度达到98.6 %。其结构经1H NM R、GC -MS 和IR 表征。
Abstract:
A homogeneo us reaction of ethy l 6 , 8 -dichlo ro -capry late and sodium sulfide takes place in aqueous methanol solution , then racemize alpha lipoic acid is o btained by hy drolysi s and acidification .The ef fect of chi ral resolut ion w as studied in to luene and at dif ferent mole ratio of ALA to PEA .The result show s that using toluene as so lvent , the amount of so lvent is 30 mL , the reaction temperature is 40 ℃, the mole ratio o f PEA to ALA is 0.5 ∶1 , the yield of R - (+) -alpha lipoic acid is 53.0 % and the optical puri ty reaches 98.6 %.The st ructure of the product w as measured by 1 H NMR , GC -MS and IR .
