摘要:
2 - 氨基- 3H - 吡咯[ 2 , 3d ] 嘧啶- 4 (7H) - 酮是一种重要的药物中间体。以氰乙酸乙酯为起始原料, 经过烷基化反 应、以及后续的环合反应合成了目标产物3 。实验结果表明, 碳酸钾作为碱对氰乙酸乙酯的去质子效果最好; 合成中间产物2 的最佳反应时间为215 h , 并且保护性气体的应用在这步合成中效果明显; 在生成最终产物的反应中, 最适宜的w (盐酸) 是 12 %。全合成产率为6.4 %。产物的结构经过核磁共振氢谱、元素分析和熔点测试得到表征。
Abstract:
2 - amino - 3H - pyrrolo [2 , 3d ] pyrimidin - 4 (7H) - one is an important p harmaceutical in2 termediate. Starting material cyanoet hyl acetate was alkylated , and followed by subsequent ring clo sures to yield t he targeting product 3. It was concluded f rom t he experiment t hat potassium carbonate as a base worked best in the deprotonation of cyanoethyl acetate ; t he optimal reaction time for the synthesis of inter2 mediate 2 was 215 hour s , and application of protective atmosp here in t his step obviously demonst rated ben2 eficial effect ; the optimal percentage concent ration of hydrochloric acid was 12 % in t he process of genera2 ting t he final product . The overall yield of the whole syntheses was 614 %. St ruct ures of t he product s were characterized by NMR spect roscopy , elementary analysis and melting point measurement .
