摘要:
以酪氨酸为原料 , 碱 性 ( N aO H) 条 件下 与溴 化苄 反应 7 h , 采用 ED TA - N a 的水 溶液 洗涤 粗品 得 O - 苄 基 酪氨 酸 (Ⅱ) , n ( O - 苄基酪氨酸) ∶ n (亚硝酸钠) ∶ n (浓硫酸) = 1 ∶ 3 ∶ 5 , 亚硝酸钠滴加时间 20 - 30 min , 反应温度 0 - 5 ℃ , 得 中间体 3 - ( 4 - 苄氧基苯基) - 2 - 羟基丙酸 ( Ⅲ) , 经 酯化 、 催化氢化脱苄 基 、 溴化反应得目 标产物 3 - ( 3 , 5 - 二溴 - 4 - 羟基苯基) - 2 - 羟基丙酸甲酯 ( Ⅵ) , 总收率 43. 6 % , 其结构经1 HN M R 和 LC - M S 表征 。
Abstract:
Starting from the D , L - ty rosine , the alky latio n by benzy l bromide and N aO H to affo rd the de- sired 3 - (4 - (benzylo xy) pheny l) - 2 - hy dro xy propanoicacid ( Ⅱ) after the w ork - up wi th aq . ED- TA - N a s olutio n . The treatment of Ⅱ wi th N aNO 2 converted the N H 2 to O H g ro up . In this s tep , n (O - benzyl - ty ro sine) ∶ n (N aN O 2) ∶ n (conc . H 2SO 4) = 1 ∶ 3 ∶ 5 , 20 - 30 min , 0 - 5 ℃ , respectively . A fter es terificatio n , Pd/ C hydrog enatio n and bromi nati on , the fi nal pro duct was o btained in an o verall y ield of 43. 6 % . The structure w as cha racterized by 1 H - N M R and LC/ M S .
